Our whole blood assay allows to measure thrombin generation (TG) in the presence of all blood cells. The assay is based on the Calibrated Automated Thrombogram (CAT) method, and utilizes α2-Macroglobulin-thrombin as the TG calibrator and tissue factor as TG trigger. The obtained TG curves provide quantitative insights into the kinetics of thrombin formation and inhibition during coagulation in whole blood.

Our plasmin generation (PG) kit is designed to monitor plasmin formation and inhibition in plasma, based on the established approach employed by calibrated automated thrombography. The assay utilizes α2-Macroglobulin-plasmin as the PG calibrator, and a combination of tissue factor, phospholipids and tissue plasminogen activator as the PG trigger. The plasmin-specific fluorogenic substrate is automatically dispensed using the Fluoroskan equipped with the Thrombinoscope software. The obtained PG curves are expressed in molar concentrations and provide quantitative insights into the kinetics of plasmin formation and inhibition during fibrinolysis.

The platelet ATP release assay measures ATP released from dense granules of platelets in platelet-rich plasma and whole blood, upon activation by a specific stimulus. The released ATP is quantified using a luciferase/luciferin reaction. This luminescence-based method offers a sensitive and precise evaluation of platelet activation and vitality.

The whole blood flow assay evaluates different aspects of the haemostatic process. This system combines microfluidics with an imaging system that operate at 37°C and employs arterial or venous shear rates. During a 10-minute perfusion, citrated blood flows over a thrombogenic surface to focus on platelet activation, aggregation and thrombus formation. In addition, a coagulant or fibrinolytic trigger can be added to study platelet-dependent coagulation, or fibrinolysis, respectively.
